The Effect of Lysogenic Induction on the Deoxyribonucleases of Escherichia Coli K12-lambda. I. Appearance of a New Exonuclease Activity.

چکیده
Infection of Escherichia coli cells with bacteriophages of the T series has been shown to result in a large rise in deoxyribonuclease activity (1, 2). Wormser and Pardee have reported that although T2 infection of E. coli K12X causes a 4to 8-fold increase in deoxyribonuclease (DNase) activity of the cells, lysogenic induction of these cells by ultraviolet light causes no significant increase in DNase activity (3). We have found that lysogenic induction of E. coli K12X by mitomycin C or by thymine deprivation (4) causes an approximately lo-fold rise in DNase specific activity observable at pH 10 in crude extracts. This elevated DNase activity is not seen when nonlysogenic cells of K12 (K12S) are treated with mitomycin C. Infection of K12S cells with a lethal mutant of X (X C-57), however, does cause a large increase in DNase activity at pH 10 in crude extracts of the infected cells. This increased DNase activity therefore seems to be specifically associated with lysogenic induction and the formation of the phage X. The DNase responsible for this rise in activity has been isolated, and evidence will be presented that this is a new exonuclease activity which is not detected in extracts of normal, uninduced E. coli K12X. Three other DNases have also been separated from lysogenically induced cells of E. coli K12X and have been found to be similar to the corresponding nucleases isolated from uninduced cells (5). A preliminary communication of this work has appeared (6).

Properties of Escherichia coli expressing bacteriophage P22 Abc (anti-RecBCD) proteins, including inhibition of Chi activity.
Escherichia coli strains bearing plasmids expressing phage P22 anti-RecBCD functions abc1 and abc2 were tested for the presence of recBC-like phenotypes. Abc2 induces moderate sensitivity to UV light in wild-type and recD mutant strains but severely sensitizes both recF and recJ mutants. Abc1 has little effect on UV sensitivity in wild-type or recF or recJ mutant hosts but increases the sensiti...
متن کاملThe Effect of Lysogenic Induction with Mitomycin C on the Deoxyribonucleic Acid Polymerase of Escherichia Coli K12-lambda.
Infection by the virulent bacteriophages of the T series is known to cause the formation of a number of new enzymatic activities in Escherichia coli (1). One of the virus-induced enzymes which appears in large amounts after T2 infection of E. coli is a new DNA polymerase which differs markedly from the DNA polymerase of normal E. coli (2). In addition, it is known that the DNA of the T phages i...
متن کاملExonuclease VII of Escherichia coli.
A new exonuclease of Escherichia coli K12, exonuclease VII, has been purified 1700-fold and characterized. The enzyme is specific for single-stranded DNA and can initiated hydrolysis at both 5' and 3' termini. It is also capable of thymine-dimer excision in vitro. The limit products of the reaction are oligonucleotides, predominantly in the range of tetramers to dodecamers. DNA is hydrolyzed b...
متن کاملEtude Au Microscope Electronique Des Transformations Nucleaires De E. Coli K12 S Et K12 S (λ26) Apres Irradiation Aux Rayons Ultraviolets Et Aux Rayons X
Nuclear transformations induced in E. coli K12S and K12S(lambda(26)) by ultraviolet radiations and x-rays have been studied with ultrathin sections in the electron microscope. The nucleoplasm keeps its normal aspect during "fragmentation" and during "condensation" of the nucleus into the "vesicular" form. Serial sections show that the "fragmented" nucleus consists in reality of only one very to...
متن کاملVaccinia DNA topoisomerase I promotes illegitimate recombination in Escherichia coli.
Vaccinia virus encapsidates a Mr 32,000 type IDNA topoisomerase. Although the vaccinia gene encoding the topoisomerase is essential for virus growth, the role of the enzyme in vivo remains unclear. In the present study, the physiologic consequences of vaccinia topoisomerase action have been examined in a heterologous system, Escherichia coli. The vaccinia topoisomerase gene was inducibly expres...
